O repertório de canções é um dos quesitos mais importantes nos games musicais. Nesse aspecto, Rock Band 2 está bem servido, contando com 84 canções dentro do disco. O gosto musical é pessoal, mas com tantas opções, são maiores as chances de encontrar canções que agradem. A lista é eclética, indo do rock progressivo do The Who, da década de 1960, até o novíssimo Shackler's Revenge, canção inédita do Guns n' Roses. São 40 anos de história do rock, passando por estilos como o heavy metal, alternativo, grunge, indie e pop-rock. Mais uma vez, a modalidade principal é a de turnê. A banda montada pelo jogador, que pode ter de um a quatro integrantes, começa pequena, tocando em "porões" nas redondezas. Mas, à medida que ganha fãs e dinheiro, além dos "acessórios" que fazem parte de uma banda de rock - vans, roadies, empresários, aviões etc. -, o grupo vai conquistando novos territórios. Desta vez, o jogador não é obrigado a escolher níveis de dificuldades mais altos para ganhar fãs, pois, agora, todos os shows realizados com sucesso acrescentam popularidade, mesmo que numa proporção menor que no começo da carreira.

Para quem quer desafio, "Rock Band 2" também tem muito para oferecer. O título traz mais canções de dificuldade elevada; nas partes finais do modo de turnê, o jogador se depara com várias músicas de alta complexidade, principalmente do gênero heavy metal.
Desta vez, o baterista ganhou mais atenção da Harmonix: agora, existe um modo de treino somente para o instrumento. No modo Beat Trainer, há 76 ritmos para praticar em várias velocidades. Também tem o Fill Trainer, em que se aprende as viradas.
Freestyle Mode, para tocar a bateria livremente. O modo de treinamento também inclui um tutorial para guitarra e uma opção para praticar as canções.
Os controles em forma de guitarra e bateria mudaram muito pouco em relação ao antecessor. imitação de Fender Stratocaster que vem em Rock Band 2 é sem fio, e traz um dispositivo que calibra automaticamente o jogo. Basta escolher a opção Calibrate e colocar o controle, primeiro, perto do alto falante e, depois, em frente à tela. A bateria também eliminou o fio, e os tambores agora tem sensores de pressão - dependendo da intensidade da batida, o volume produzido varia em três níveis -, mas não são muito mais silenciosos que antes. O pedal foi reforçado com partes metálicas, o que deixa aliviado quem tem um estilo mais agressivo de tocar.
